Overview:

Calcium antagonists are widely used to treat angina, systemic hypertension, and other cardiovascular diseases. 1,4-dihydropyridine (DHP) calcium antagonists bind to specific receptors in cardiac and smooth muscles, thereby inhibiting the influx of extracellular calcium when these muscles are stimulated to contract. The prototype drug of DHP, nifedipine, only has short-acting cardiovascular effects. Later, some second- and third-generation DHP drugs with long-acting antihypertensive effects appeared.
